目的从鼠肺中获得汉坦病毒分离株并进行基因鉴定与分型,为地区分离株特性研究提供基础。方法用Vero-E6细胞分离病毒,进行荧光和RT-PCR鉴定,之后对目的片段进行序列测定和分析。结果从褐家鼠鼠肺中分离到一株汉坦病毒,编号SC106。用汉城型(SEO)汉坦病毒M基因特异性引物检测得到预计大小的片段,此片段的核苷酸序列分析表明,SC106与SEO型参考毒株的同源性较高,均在94%以上;与汉滩型(HTN)参考毒株的同源性较低,均在67%以下。进化树分析表明SC106位于SEO型所在的支系。结论SC106为SEO型汉坦病毒。
OBJECTIVE: To obtain Hantavirus isolates from rat lungs and to identify and classify them, providing the basis for the study of the characteristics of regional isolates. Methods Vero-E6 cells were used for virus isolation and identification by fluorescence and RT-PCR. The sequence of the target fragment was determined and analyzed. Results A strain of Hantavirus was isolated from the brown house mouse rat lung, number SC106. The predicted size of the fragment was detected with Seoul National (Hantavirus) Hantavirus M gene-specific primers. The nucleotide sequence analysis of this fragment showed that the homology between SC106 and SEO reference strains was higher than 94% Homologous to Hantaan-type (HTN) reference strains was lower than 67%. Phylogenetic tree analysis shows that SC106 is located in the domain of SEO type. Conclusion SC106 is SEO type Hantavirus.
