目的 观察博莱霉素 (bleomycin ,BLM)所致小鼠肺损伤后纤维化时广谱钙粘附素 (pan cadherin,Pan cd )及 β连环素 (β catenin ,β cat)表达的动态变化 ,探讨钙粘附素、连环素表达与肺纤维化发生发展的关系。方法 小鼠气管内注入BLM复制肺纤维化模型。分期处死动物 ,以抗Pan cd及抗β cat抗体行肺组织免疫组织化学染色。 结果 肺纤维化时 ,Ⅰ型肺泡细胞Pan cd表达下调 ,Ⅱ型肺泡细胞被新诱导表达Pan cd。随后 ,细支气管上皮Pan cd与 β cat的表达均下调 ,并呈现出下调 恢复 下调的动态变化 ,同时伴Pan cd表达由胞膜型转变为胞浆型 ,且呈现出胞浆型 胞膜型 胞浆型表达的动态变化。肺泡巨噬细胞 (AM)及肺间质细胞Pan cd表达增加。结论 Pan cd、β cat的表达异常参与了博莱霉素致小鼠肺组织损伤后纤维化的形成
Objective To observe the dynamic changes of the expression of pan cadherin (Pan cd) and β catenin (β cat) after bleomycin (BLM) induced lung injury in mice, To investigate the relationship between cadherin, catenin expression and pulmonary fibrosis. Methods The mice were injected intratracheally with BLM to induce pulmonary fibrosis. Animals were sacrificed by staging and immunohistochemical staining of lung tissue was performed with anti-Pan cd and anti-β cat antibodies. Results Pulmonary fibrosis, Panc-1 alveolar cells down-regulated expression of type II alveolar cells were induced by the new expression of Pan cd. Subsequently, the expression of Pan cd and β cat in the bronchiolar epithelium was down-regulated and showed a down-regulation and down-regulation dynamic change. At the same time, the expression of Pan cd was changed from cytoplasmic to cytoplasmic and showed cytoplasmic cell Dynamic changes in plasma expression. Alveolar macrophages (AM) and pulmonary interstitial cells Pan cd expression increased. Conclusions The abnormal expression of Pan cd and β cat is involved in the formation of fibrosis induced by bleomycin-induced lung injury in mice
