目的:观察球囊损伤对动脉平滑肌细胞(SMC)表皮生长因子受体(EGFR)表达的影响,探讨EGFR与再狭窄的关系。方法:于兔右颈总动脉球囊损伤前及损伤后24h、48h、72h、7天和14天,取实验动脉段用免疫组化LSAB法检测EGFR的表达。结果:正常颈总动脉中膜未见EGFR阳性表达,动脉球囊损伤后24h中膜部分SMC显示EGFR阳性表达,48h中膜呈EGFR阳性表达的SMC较24h增加,72h又较48h进一步增加;7天和14天动脉中膜EGFR阳性表达的SMC均较72h显著减少,但新生内膜中增殖的SMC显示出大量的EGFR阳性表达。阳性染色主要定位于SMC的胞膜。结论:球囊损伤使动脉EGFR表达显著增加,EGFR与再狭窄形成过程中SMC增殖有关。
OBJECTIVE: To observe the effect of balloon injury on the expression of epidermal growth factor receptor (EGFR) in arterial smooth muscle cells (SMC) and to explore the relationship between EGFR and restenosis. Methods: The experimental arterial segments were examined by immunohistochemistry (LSAB) before and at the right carotid artery balloon injury and at 24h, 48h, 72h, 7d and 14d after injury. Results: The expression of EGFR in normal carotid artery was not observed in normal carotid artery. The expression of EGFR in SMMC of middle part of carotid artery 24 hours after arterial balloon injury was higher than that in 24h and 48 hours after injury. The SMC of positive expression of EGFR on day and on day 14 was significantly lower than that on 72h, but the proliferation of SMC on neointimal showed a large number of positive EGFR expression. Positive staining mainly located in the cell membrane of SMC. CONCLUSION: Balloon injury significantly increases the expression of EGFR in arteries and EGFR is related to the proliferation of SMC during restenosis.
